چکیده

Forward genetic analysis is one of the principal advantages of the zebrafish model system. However, managing zebrafish mutant lines derived from mutagenesis screens and mapping the corresponding mutations and integrating them into the larger collection of mutations remain arduous tasks. To simplify and focus these endeavors, we developed an approach that facilitates the rapid mapping of new zebrafish mutations as they are generated through mutagenesis screens. We selected a minimal panel of 149 simple sequence length polymorphism markers for a first-pass genome scan in crosses involving C32 and SJD inbred lines. We also conducted a small chemical mutagenesis screen that identified several new mutations affecting zebrafish embryonic melanocyte development. Using our first-pass marker panel in bulked-segregant analysis, we were able to identify the genetic map positions of these mutations as they were isolated in our screen. Rapid mapping of the mutations facilitated stock management, helped direct allelism tests, and should accelerate identification of the affected genes. These results demonstrate the efficacy of coupling mutagenesis screens with genetic mapping.

Zebrafish: genomics on the fast track.

The zebrafish (Danio rerio) has proven its worth as an excellent model system for the study of vertebrate development. In 1996, in an issue of the journal Development, the results of the characterization of almost 2000 mutations derived from two screens of ENU-mutagenized zebrafish (Driever et al. 1996; Haffter et al. 1996) were reported in 36 articles (Vol. 123, Dec. 1996). The same issue, how...

From screens to genes: prospects for insertional mutagenesis in zebrafish.

The systematic, and now classic, genetic screens for mutations affecting the development of Drosophila and Caenorhabditis elegans have laid the foundation for the study of developmental mechanisms in these invertebrates (N/isslein-Volhard and Wieschaus 1980; Horvitz and Sternberg 1991). The subsequent molecular isolation and characterization of the affected genes has allowed a thorough understa...

Molecular dissection of craniofacial development using zebrafish.

The zebrafish, Danio rerio, is a small, freshwater teleost that only began to be used as a vertebrate genetic model by the late George Streisinger in the early 1980s. The strengths of the zebrafish complement genetic studies in mice and embryological studies in avians. Its advantages include high fecundity, externally fertilized eggs and transparent embryos that can be easily manipulated, inexp...

Rapid identification of kidney cyst mutations by whole exome sequencing in zebrafish.

Forward genetic approaches in zebrafish have provided invaluable information about developmental processes. However, the relative difficulty of mapping and isolating mutations has limited the number of new genetic screens. Recent improvements in the annotation of the zebrafish genome coupled to a reduction in sequencing costs prompted the development of whole genome and RNA sequencing approache...

The zebrafish model system in cardiovascular research: A tiny fish with mighty prospects

1. INTRODUCTION The zebrafish Danio rerio, a tropical freshwater fish, belongs to the family of cyprinidae, which in the last 30 years has developed into a very popular model organism for studies of embryonic development and human diseases. Initially the zebrafish species has been selected on the basis of its small size of approximately 3–5 cm, its transparency during development and its high f...
